Department of Genetic Toxicology and Nanotoxicology (DGTN) is focused on the mechanisms of toxic effects of engineered nanoparticles as well as combustion generated fine and ultrafine particles. Major attention is paid to the interaction of particles and their components with biomolecules inhuman cell lines. The methods include various tests of cytotoxicity, the detection of reactive oxygen species and oxidative damage of DNA, proteins and lipids (ELISA), analyzes of genotoxicity (Comet assay, DNA adducts) and clastogenic effects (micronucleus test and cytogenetic analysis by FISH),as well as analyzes of protein expression (western blotting) and DNA repair activity (NER and NHEJ). To get more insight into the mechanism of biological effects of particles or their extracts, the toxicogenomic methods (microarrays and next generation sequencing) and RT-PCR are used. Most ofthese tests are also available to external users from research, universities and academia via the Research Infrastructure NanoEnviCz(www.nanoenvicz.cz).DGTN also concentrates on the mechanisms of adverse effects of engine emissions particularly under real traffic condition (www.medetox.cz). Another topic includes molecular epidemiology studies analyzing the effect of environmental pollution on human health.

Important result in 2015



Analysis of genetic damage in lymphocytes of former uranium processing workers.
The frequency of cells containing micronuclei (MN) and the presence of centromeres in the MN were analyzed in lymphocytes of men from Southern Bohemia (former workers of a former uranium processing plant „MAPE Mydlovary“ and controls). No differences were found between formerly exposed workers and the control group. Moreover former workers with X-ray examination had significantly lower level of DNA damage than the control group. Possible reason of this results may be adaptation of the organism.

2. Day-to-day variability of toxic events induced by organic compounds bound to size segregated atmospheric aerosol.
The temporal variability of size-segregated aerosol mass collected on a daily basis during the winter period in highly polluted districts of Ostrava city as well as toxic effects of organic compounds extracted from each size fraction were quantified. We revealed that upper accumulation mode fraction (0.5 < dae < 1 μm) comprises most of the aerosol mass and contained 44% of total c-PAHs while ultrafine fraction (< 0.17 μm) contained only 11% of c-PAHs. DNA adduct levels and dioxin-like activity strongly correlated with both aerosol mass and c-PAH concentrations suggesting genotoxicity as a major toxic effect of organic compounds bound to size-segregated aerosol.

3. Reduced gene expression levels after chronic exposure to high concenrations of air pollutants.
We analyzed the effect of air pollution on gene expression in subjects living in Prague and in the polluted Ostrava region. We expected to observe changes in expression of DNA repair genes in subjects from the Ostrava region. Unexpectedly, the changes were found in Prague subjects, particularly in genes associated with immune response and neurodegenerative diseases. The results suggest that chronic exposure to air pollution may result in adaptation of the organism with possible negative health effects.

Important result in 2014



1. Preterm birth, infant weight gain, and childhood asthma risk: A meta-analysis of 147,000 European children
Younger gestational age at birth and higher infant weight gain were independently associated with higher risks of preschool wheezing and school-age asthma. Compared with term-born children with normal infant weight gain, we observed the highest risks of school-age asthma in children born preterm with high infant wight gain (OR 4.47; 95% CI, 2.58-7.76).

2. Analysis of gene expression changes in A549 cells induced by organic compounds from respirable air particles
We studied a toxic effects caused by the organic compounds from respirable air particels (PM2.5) in human lung A549 cells. Gene expression analysis revealed numerous deregulated genes and pathways related to activation of the aryl hydrocarbon receptor (AhR), metabolism of xenobiotics and endogenous compounds. We further identified deregulated genes involved in pro-inflammatory processes, oxidative stress response and in cancer and developmental pathways, such as TGF-β and Wnt signaling pathways. Interestingly, no DNA damage response was identified.

 
Venn diagrams representing numbers of common and locality-specific deregulated genes following 24-h treatment of A549 cells by 3 subtoxic concentrations of extractable organic matters (EOMs). EOMs were prepared by organic extraction of particulate matter <2.5 μm (PM2.5) collected in 4 localities of the Czech Republic differing by the extent and sources of the air pollution: Ostrava-Bartovice, Ostrava-Poruba, Karvina and Trebon. A549 cells were treated with the EOMs and total RNA from A549 was isolated. Global gene expression analysis was performed using whole genome Illumina chips as described in Section 2. The intersections represent the number of genes deregulated simultaneously at various localities relative to DMSO used as a solvent control (adjusted P-value < 0.05, average expression level > 4). The overlap of deregulated genes among the localities at the following concentrations: 10 μg/ml (A), 30 μg/ml (B), 60 μg/ml (C).

3. Nonhomologous DNA end joining and chromosome aberrations in human embryonic lung fibroblasts treated with environmental pollutants.
We studied the effect of environmental pollutants (carcinogenic benzo[a]pyrene (B[a]P) and organic extracts from particulate matter (PM2.5)) on DNA damage and repair. We analyzed key steps of DNA damage, including the detection of DNA breaks, DNA repair proteins, ligation (joining) of DNA ends and induction of chromosome aberrations. We found that tested compounds, particularly organic extracts, induce DNA damage followed by its repair. This, however, results in the induction of chromosome aberrations.

2. Ultrafine particles are not major carriers of carcinogenic PAHs and their genotoxicity in size-segregated aerosols.
The ultrafine aerosol fraction is hypothesized to be the most important carrier of carcinogenic-PAHs, since it possesses the highest specific surface area of PM. However, we demonstrated that the fraction of ultrafine particles of various ambient-air samples is neither a major carrier of c-PAHs, nor a major inducer of their genotoxicity, which is an important finding that is relevant to the toxicity and health effects of ultrafine particles, which are so extensively discussed these days.

3. Air pollution affects methylation profiles in the pediatric population.
Gene activity is significantly influenced by their methylation. On the settings for each individual has influence mainly the negative effects of exposure during the prenatal period. In our study, we compared the distribution of 27,578 methylated sites for groups of children of the city and Prachaticko. Children of the city, where the concentration of carcinogenic benzo [a] pyrene (B [a] P) was significantly higher methylation had different distribution groups, suggesting a different gene activity due to air pollution.

 

Methylation of genes in the children of the city is different from the children of Prachaticko. Adaptive response in subjects exposed to polluted air? Despite exposure to high concentrations of air pollutants subjects living in the Ostrava region exhibited changes of expression in lower number of genes than Prague inhabitants. The result suggests adaptation of the human organism to adverse environmental conditions.
